Ok.. I isolated my clones from the agar plates yesterday. Before the overnight culture, the broth is clear:
After 18 hrs at 37 C (body temperature) in the shaking incubator:
We get turbidity. (It means they grew). You can actually measure the colony conc. based on this turbidity.
For now.. I could either go straight to plasmid isolation (procedure will take ~ 4 hrs with this many samples) or, they go in the fridge and i do it monday. Fridge will slow down the growth so they won't overgrow and die and split open. If that happens I lose the little plasmids they are carrying inside them. And I need those. Night Night little clones!
